Requirements:
The candidate should have PhD in biology (specialization in endocrinology of reproduction and andrology)

Conditions of employment: good knowledge of physiology and endocrinology of male reproductive system, practical knowledge of histological techniques, gene expression analysis techniques (e.g. RT-PCR and real-time RT-PCR) and proteins (e.g. Western blot, immunohistochemistry / immunofluorescence radioimmunoassay techniques, cytological analyzes, laboratory experience and experience in work with laboratory animals (in vivo experiments, tissue isolation), practical knowledge of ex vivo tissue culture techniques and male gonadal cells in vitro (Leydig cells: primary cultures and cell lines), spoken and written Polish and English.

Description of tasks:
Post-doc will participate in the project: Research on hormonal and molecular mechanisms of tumor development in interstitial male gonadal tissue funded by the National Science Center (OPUS12) for 2017-2020. The main objective of the project is to determine the contribution of signaling with GPER and PPAR receptors in the growth, proliferation and lipid homeostasis in Leydig cells as well as in the development of Leydigioma. Tasks:

1. Characterization of GEPR and PPAR expressions.

2. Determination of the role of GPER and PPAR-in vitro.

3. Verification of identified GPER- and PPAR- regulated genes and their products related to growth, proliferation, and lipid homeostasis – in vitro and ex vivo study.

4. Determination of the effect of mixture of endocrine disrupters and sex hormones on expression of GPER and PPAR- regulated genes and their products related to growth, proliferation, and lipid homeostasis- in vitro and in vivo study.
5. Determination of the relationship between expression of GPER- and PPAR-regulated genes and Leydig cell morphological and biochemical characteristics – in vitro and ex vivo study.

Specific tasks:
Performing of qRT-PCR analyses, preparation of material for analysis of microarray (tasks 1,2); performing of transfection of siRNA (task 2); verification of the identified genes (tasks 1-3); performing of immunoassays and radioimmunoassays and cytometric analyses, performing of a part of co-immunoprecypitation assays (tasks 2-4); Participation in tasks 3-4: preparation of human and mouse tissues for molecular analyses; performing of cell signaling assays; performing of a part of WB analyses; performing of proliferation, apoptosis, migration and angiogenesis assays; analysis and preparation of results for publication.
